Overview
Cells can be cultured using CytoSoft®-Imaging products on PDMS substrates with rigidities of 0.2, 0.5, 2, 8, 16, 32, and 64 kPa. One glass-bottom 24-well plate with a glass bottom is included with this CytoSoft®-Imaging plate for high-resolution cell imaging on the chosen stiffness.
The 24-well products from CytoSoft®-Imaging have a #1.5 glass bottom that is attached to a 24-well plate and a defined elastic modulus. Each well has a layer of silicone that is about 0.03-mm thick and has a uniform thickness of silicone gel.
Before the incorporation of cells, the silicone gels are activated and prepared to bind to an ECM that has been purified, such as PureCol® type I collagen (#5005).

Features
- Cell morphology and gene expression can be significantly impacted by the substrate’s rigidity, to which cells cling
- Products from CytoSoft® offer a method for cultivating cells on substrates of different rigidities to accommodate a wide range of physiological conditions
- If low autofluorescence and exceptional optical clarity are necessary for high-resolution imaging, use the CytoSoft® - Imaging plate
- The plate has a black polystyrene frame attached to a #1.5 glass bottom plate, and it also has a lid
- There is one plate per package of the plates, which have been sterilized using ozone
- A thin layer of specially formulated biocompatible silicone with an elastic modulus (rigidity) that has been meticulously measured and certified covers the bottom of each well
- In CytoSoft® products, the gel surfaces are functionalized to create covalent bonds with protein amines
- The gel surfaces can be coated with matrix proteins and cells can be plated more easily because the chemical functionalization is stable and the reaction does not need a catalyst
Applications
- The silicone substrates have almost no auto-florescence and are optically clear. Each well's silicone layer is tightly attached to the well’s base
- In contrast to hydrogels (like polyacrylamide gels), which are prone to hydrolysis, silicone gels are resilient and resistant to tearing or cracking, and their elastic moduli (rigidities) remain largely unchanged during prolonged storage times
- They also do not dry out or swell
- Products from CytoSoft®-Imaging support live cell staining with membrane and cell-permeable dyes, cell fixation with widely used methods like paraformaldehyde, and immunostaining of fixed cells
